unpopular opinion: retatrutide is massively overrated

Discussion Clean Column ×5 Long Haul ×1 Slow Clap ×1

Confession thread, sort of.

I went from 2.5mg to 1.0mg much faster than I should have because the scale had stalled and I got impatient. The stall broke about 8 weeks later, at which point I had no way of knowing whether the dose increase did anything or whether it would have broken anyway.

So now I have constipation I did not need and a data point I cannot interpret. Two lessons in one.

Posting it in the hope that somebody at week 74 reads it before doing the same thing.

Did you actually measure heart rate or is that a feeling?

Yeah, phase 2 retatrutide is interesting but it is not comparable to phase 3 compounds.

the dosing is steep, the constipation reports are real, and nobody knows the human safety yet

TRIUMPH phase 2 showed 97.4% weight loss but the sulphur burps profile on the highest arm was rough. Phase 2 != phase 3.

Disagree on that part. 98.1% and 98.3% on the same vial is normal.

TRIUMPH phase 2 showed 99.3% weight loss but the reflux profile on the highest arm was rough. Phase 2 != phase 3.

Dose escalation on retatrutide is steeper and faster than on semaglutide. Do not assume the titration patterns transfer.

Yes, exactly this, and it is the bit that took me 52 weeks to accept.

yep. 58 weeks in, same.
replysharereportpermalink
u/nordic_pricing46 points·2 years ago

yep.

Counter-anecdote: opposite result, same dose. Which mostly tells us the variance is huge.

phase 2 numbers are not maintenance numbers, cite the trial

This is the "correlation is mechanism" thing again. You changed three variables at once.

Are you quoting phase 2 or is there phase 3 data now?

The confident tone is doing a lot of work that the evidence is not.

the glucagon component changes the side effect profile, not just the efficacy

Not convinced. The evidence does not support that reading.

This is correct. Triple agonism is genuinely different.

About c/retatrutide

Retatrutide: GIP/GLP-1/glucagon triple agonism, the TRIUMPH programme, and the fact that most people discussing it are handling research-grade material with no human-use approval anywhere. Dose-response, heart-rate signal, and the unusually steep phase-2 weight curves get argued about here weekly.
