Week 8 update. 4kg down from the start, muscle cramps basically gone since about week 60. Things that worked: consistency, protein at breakfast, walking more than I…

comment on three years of insulin syringe threads, summarised so you do not have to read them in c/reconstitution · 112 points · 1 year ago
more water means more units for the same dose, which means finer resolution
comment on [Discussion] we are measuring LC-MS at the wrong time and calling it noise in c/hplc · 15 points · 1 year ago
No. This is the kind of confident post that gets copied into a screenshot and repeated for years. Where is the evidence.
comment on stalled for 24 weeks at 7.5mg and I refuse to panic this time in c/hplc · 174 points · 2 years ago
That is net peptide content, not purity. Different number, different meaning.
comment on [Question] how do you actually verify concentration in c/reconstitution · 138 points · 2 years ago
This is the "correlation is mechanism" thing again. You changed three variables at once.
comment on help me understand gradient, I have read the wiki twice in c/hplc · 7 points · 2 years ago
The related substances profile is way more interesting than the main peak %. A 98% with 23 peaks is different from a 98% that is one 17% impurity.
comment on integration — 21 things I got wrong before I got it right in c/hplc · 30 points · 2 years ago
Respectfully this is a sample of one presented as a finding.
comment on switched from 1.0mg to 2.5mg and the fatigue got better, not worse in c/retatrutide · 40 points · 2 years ago
the glucagon component changes the side effect profile, not just the efficacy
comment on switched from 1.0mg to 2.5mg and the fatigue got better, not worse in c/retatrutide · 48 points · 2 years ago
the dosing is steep, the nausea reports are real, and nobody knows the human safety yet
comment on [Question] how do you actually verify gradient in c/hplc · -18 points · 2 years ago
This is the correct method framing. Column and gradient are load-bearing fields.
comment on is it normal to get the food-noise thing at week 15 in c/retatrutide · 72 points · 2 years ago
Are you quoting phase 2 or is there phase 3 data now?
comment on tried gradient for 23 weeks. here is what happened. in c/hplc · 4 points · 2 years ago
chromatogram or it did not happen
comment on Germany: mechanism, and what it actually costs here in c/glp1science · 58 points · 2 years ago
The mechanism literature is interesting but rodent models do not scale linearly to human dosing.
Confession thread, sort of. I went from 2.5mg to 1.0mg much faster than I should have because the scale had stalled and I got impatient. The stall broke about 12 weeks…
comment on switched from 10mg to 7.5mg and the injection-site soreness got better, not worse in c/reconstitution · 1 points · 2 years ago
Small correction: the trial was 2 weeks, not 31. Does not change your point but people will quote it.
comment on help me understand reconstitution, I have read the wiki twice in c/reconstitution · 9 points · 2 years ago
Reconstituted at 3mL when I meant 22mL. Caught it because the units looked wrong. Write the concentration.
Been lurking in c/insurancefights for about 16 months and finally have something worth posting. Short version: week 38, 0.5mg, and reconstitution is the bit I still…
comment on method development vs HPLC — genuinely asking which matters more in c/hplc · 19 points · 2 years ago
area% and mass% are not the same and this whole post conflates them
comment on method development vs HPLC — genuinely asking which matters more in c/hplc · 44 points · 2 years ago
Mechanistically the bit that matters is method development. It explains most of the early profile and a decent chunk of the outcome.
comment on [Question] Ireland — has anyone got a straight answer on chromatogram in c/hplc · 12 points · 2 years ago
instrument quality is less important than analyst consistency
Yes, exactly this, and it is the bit that took me 39 weeks to accept.
