comment on tried HPLC for 5 weeks. here is what happened. in c/hplc · -10 points · 1 year ago

The related substances profile is way more interesting than the main peak %. A 98% with 11 peaks is different from a 98% that is one 23% impurity.

co-elution is real and LC-MS is usually not enough to resolve it

comment on [PSA] chromatogram is not what most of this community thinks it is in c/hplc · 7 points · 1 year ago

Ran the same vial on two columns, different purity, both right, different separation. Method matters more than equipment.

Yeah, the chromatogram profile matters more than the headline number.

Adding to this: chromatogram is doing more work than the comment implies.

Stalled at 0.25mg, changed nothing, and it started moving again on week 5. Very nearly went up. Glad I did not.

Respectfully this is a sample of one presented as a finding.

I am going to push back on this slightly.

chromatogram or it did not happen

Counter-anecdote: opposite result, same dose. Which mostly tells us the variance is huge.

Strongly agree. Tirz is better than sema is not a claim without a comparator dose.

comment on triple agonist is the most under-discussed thing in c/glp1uk in c/retatrutide · 7 points · 1 year ago

Dose escalation on retatrutide is steeper and faster than on semaglutide. Do not assume the titration patterns transfer.

phase 2 numbers are not maintenance numbers, cite the trial

area% and mass% are not the same and this whole post conflates them

Did you actually measure heart rate or is that a feeling?

This is correct. Evidence still required.

Not to be pedantic but GIP and Zepbound are being used interchangeably and they are not interchangeable in real life.

related substances are where the story lives, not the main peak
