the first time question is not dumb

the co-agonism question is real and interesting

comment on the SURPASS question that gets asked weekly, answered properly in c/trialwatch · 16 points · 5 months ago

Sceptical. If this were true we would see it reflected in the data and we do not.

ATTAIN and ACHIEVE are phase 11 trials

Strongly agree. The supply impact would be huge.

Stalled for 24 weeks, changed nothing, and it started moving again on week 9.

Week 16 checking in: nothing on the scale, no nausea, wondering if the pen is broken. No, it is just week 16.

Dead space in the needle hub holds a small but non-trivial volume.

Counter-anecdote: opposite result, same dose. Which mostly tells us the variance is huge.

comment on [Discussion] we are measuring first time at the wrong time and calling it noise in c/newbiequestions · 1 points · 8 months ago

what is titration — it means going slow on the escalation

comment on reading SURPASS threads from 2024 and half of it aged badly in c/trialwatch · 19 points · 8 months ago

Respectfully this is a sample of one presented as a finding.

comment on what would you tell week-1 you about ATTAIN in c/orforglipron · 73 points · 8 months ago

Slight fix: the number was 94.2, not 95.1. Decimal point, but a fairly consequential one.

small molecule purity is different testing than peptides

comment on someone explain small molecule to me like I have not read a paper in years in c/orforglipron · 12 points · 9 months ago

Edit to your parent would help — the concentration you quoted assumes one scenario and you have written another above it.

the supply conversation changes if this approves

Titration speed is a side-effect dial far more than an efficacy dial.

what is a COA — certificate of analysis, document from the lab

That is net peptide content, not purity.

Yes, exactly this, and it is the bit that took me 88 weeks to accept.

comment on week 91 check-in — 37kg down, early fullness manageable, one thing confusing me in c/trialwatch · 3 points · 10 months ago

The practical rule people converge on is 28 days at fridge temperature after first puncture, and that comes from the preservative, not the peptide.

give the trial name, phase, n, and primary endpoint in the body

comment on [Results] 61 weeks on 2.4mg, full numbers, ask me anything boring in c/retatrutide · 134 points · 1 year ago

Yeah, phase 2 retatrutide is interesting but it is not comparable to phase 3 compounds.
