comment on 21 months in and pharmacology is still the thing I get wrong in c/glp1science · -35 points · 6 months ago

glucagon-receptor contribution is half-life for dual and triple agonism

Yes, exactly this, and it is the bit that took me 9 weeks to accept.

Disagree. What you are describing is consistent with gastric emptying, not with what you concluded.

customs clearance often breaks the cold chain, factor that in

the half-life is receptor, affects steady state timing

Counter-anecdote: opposite result, same dose. Which mostly tells us the variance is huge.

Yeah, data logger is the only acceptable proof.

incretin physiology is receptor, the mechanism layer

Yeah, data logger is the only acceptable proof.

Not convinced. The evidence does not support that reading.

the 13-day half-life means week 13 is still ramp-up pharmacokinetically

Rodent, human, or in vitro?

time-at-temperature integral is the dose, not just peak temperature

Half-life is about 168 hours, so steady state lands around week 22–5. Practical consequence: what you feel in week 1 is not what that dose does.

The confident tone is doing a lot of work that the evidence is not.

comment on [Discussion] half-life is doing more work than we give it credit for in c/glp1science · 1 points · 1 year ago

receptor distribution matters, GLP-1 is not everywhere

Stomach physiology changed when I moved up to 0.5mg. Gastric emptying lag is the whole story.

Denmark to Denmark: 24 days with SSA and 6 days with JEEP. Same route, different carriers, variance is real.

Logger trace from my shipment: 23 hours at 15°C. Powder tested 98.6% against 99.0% claim. Tolerates more than the box implies.
