Twice-weekly splitting has a pharmacokinetic rationale and almost no outcome data.

comment on [Discussion] why did splitting/microdosing culture even take off here given the total lack of trial data in c/glp1science · 96 points · 2 days ago
I'd add: communities like this reward detailed, specific posts (logs, spreadsheets, worked math) over vague ones, and splitting/microdosing produces great content for that format — lots of granular numbers to post about. Standard weekly dosing just doesn't generate as much postable detail.
comment on the actual pharmacokinetic case for twice-weekly splitting, and what it doesn’t prove in c/glp1science · 231 points · 3 days ago
this is exactly my position and I'm glad someone wrote the PK case out properly instead of me just saying "no data" for the hundredth time. the mechanism is plausible, the outcome claim is not evidenced.
comment on [Explainer] half-life is 168 hours. that is why your injection day barely matters. in c/glp1science · 96 points · 3 days ago
and it is the reason twice-weekly splitting has a plausible rationale and almost no outcome data. with a week-long half-life the PK argument for splitting is weak to begin with.
comment on anyone actually tracked their own sides before/after switching to a split schedule in c/semaglutide · 143 points · 3 days ago
this is the right way to ask this question honestly, most posts just assert it helped. self-tracked before/after is still n=1 with no blinding, but it's a lot better than a vibe.

Half-life is about 168 hours, so steady state lands around week 20–5. Practical consequence: what you feel in week 1 is not what that dose does.

Not to be pedantic but mechanism and gastric emptying are being used interchangeably and they are not interchangeable in real life.
