They agree to within a point or two, which is enough to catch a 94% sold as 99% — which is the actual thing we use them for. Precision and usefulness are different requirements.

The analytical half of this is a bigger change than people assume. Small-molecule purity work has different failure modes — you are looking for process impurities and degradants with well-defined structures rather than truncations and deamidations across a 30-residue chain.

It is also, generally, easier and cheaper to characterise. Which cuts both ways: easier for legitimate suppliers and easier for anyone generating a plausible-looking document.

Stickying the c/hplc crosspost of this. One addition from the analytical side: area% is also blind to identity. A closely related impurity that co-elutes with the main peak is counted as product, which is the entire argument for LC-MS confirmation on anything unusual.

Correct flair, correct restraint in not naming, and this is exactly what the Correction flair is for — it is not a removal, it is a note.

Rule of thumb for anyone calibrating: if your smallest reported impurity is within about 3× of your baseline noise, you are reporting noise. One decimal place on a purity figure is almost always as far as the data goes.

Agreed on the trace, and thank you for separating "nice chemistry" from "good vendor". Those get conflated constantly and they are genuinely independent variables.

Also worth flagging for anyone new: a clean trace is partly the sample and partly the method. A bad method can make good material look messy and a lazy one can hide real impurities under a wide peak.

worth noting SUSTAIN 6 and the STEP trials used faster escalation than most of this "low and slow" folk wisdom and still had meaningful discontinuation from GI side effects. the label pace isn't invented, it's just aggressive.

The 3.2% single impurity is the whole question and MS can probably tell you what it is. If it is a related substance — a truncation, an oxidation, a des-amido — that is a synthesis and purification story and it is disappointing rather than alarming.

If it is unrelated, that is a different conversation. Can you get a mass on that peak?

This is the best kind of post in this community — same method, same day, one variable, and the finding is in the profile rather than the headline.

For anyone reading who is about to say "0.7 points is nothing": correct, and irrelevant. The author explicitly says so. The claim is about peak count and largest single impurity, which are different measurements that happen to appear on the same trace.
