comment on FLOW got published and nobody in here is talking about eGFR slope in c/trialwatch · 96 points · 1 days ago

watch where the curves separate too. early separation on a kidney endpoint is more surprising than early separation on weight.

Retitled to remove editorialising.

Disagree on that part. 99.2% and 96.2% on the same vial is normal.

and the regain curve over time matters more than a single endpoint number too, where the curve is steepest tells you more than the final value alone

Second this question. I have wondered about tendon for 23 months and never seen a straight answer.

same lesson as watching a survival curve, the smooth published line and any one patient's actual jagged path are almost different objects

PDF metadata said the COA was created in a browser four months after the date printed on the page. Worth flagging.

Disagree but this is the good kind of wrong — it is specific enough to be checked.

This is evidence. The metadata is checkable and it is odd.

comment on [Titration] stayed at 0.5 for eleven weeks and it was the right call in c/semaglutide · 0 points · 18 days ago

Half-life is about 168 hours, so steady state lands around week 14–5. Practical consequence: what you feel in week 1 is not what that dose does.

comment on is it bad that i am not nauseous at all in c/newbiequestions · 22 points · 18 days ago

Mechanistically the bit that matters is normal. It explains most of the early profile and a decent chunk of the outcome.

Yeah, rodent data does not scale to humans.

Ireland here. Waited 21 months on an NHS list, gave up, went private, and the cost was half what I expected but the admin was worse.
