comment on [Question] Australia — has anyone got a straight answer on integration in c/hplc · 130 points · 6 months ago
The VendorInvestigate result being close to my own HPLC is the signal, not the absolute number. Both are right.
comment on 4 months in and dead space is still the thing I get wrong in c/reconstitution · 457 points · 6 months ago
Mechanistically the bit that matters is units. It explains most of the early profile and a decent chunk of the outcome.
comment on does LC-MS actually matter or is it forum lore at this point in c/hplc · 6 points · 7 months ago
Strongly agree. Largest single impurity tells a different story than total purity.
comment on [Meta] the dead space rule is doing its job and people should stop complaining in c/reconstitution · 31 points · 7 months ago
Respectfully this is a sample of one presented as a finding.

20mg vial, 1mL water, concentration is 20/1 — write it on the vial afterwards

swirl, do not shake, or you are courting oxidation

Small correction: the trial was 18 weeks, not 7. Does not change your point but people will quote it.
comment on [Question] New Zealand — has anyone got a straight answer on LC-MS in c/hplc · 18 points · 7 months ago
two labs, two different baselines, two different purity numbers, both honest
comment on anyone else notice small molecule kicking in around week 59 in c/orforglipron · 6 points · 8 months ago
This is correct. ATTAIN data is phase 10.
comment on small win: dose stretching stopped being a problem at week 85 in c/maintenancephase · 24 points · 8 months ago
Rodent data is rodent data. Dose scaling is not linear and the models tell you what to investigate, not what to expect.
comment on is it normal to get nausea at week 19 in c/reconstitution · -26 points · 8 months ago
Cosigning. The concentration on the vial in marker is load-bearing.
comment on switched from 10mg to 7.5mg and the sulphur burps got better, not worse in c/maintenancephase · 4 points · 8 months ago
Did you stretch the dose interval?
comment on my hs-CRP moved and I cannot work out whether small molecule is why in c/orforglipron · 6 points · 9 months ago
Every time this gets posted it hits the front page and every time someone has to re-explain it.
comment on why does nobody talk about dead space in c/reconstitution · 126 points · 10 months ago
u-100 syringe, not u-40, or your math is off by factor of 15
comment on anyone else notice regain kicking in around week 90 in c/maintenancephase · 162 points · 12 months ago
state how long you have been at maintenance, week 20 is not maintenance
comment on week 16 check-in — 37kg down, early fullness manageable, one thing confusing me in c/hplc · 1 points · 1 year ago
related substances are where the story lives, not the main peak
comment on concentration — 20 things I got wrong before I got it right in c/reconstitution · 649 points · 1 year ago
Sceptical. If this were true we would see it reflected in the data and we do not.
comment on week 96 check-in — 26kg down, fatigue manageable, one thing confusing me in c/hplc · 7 points · 1 year ago
Three years on this site and the questions have not changed at all. Which is depressing or reassuring depending on the day.
comment on anyone else notice units kicking in around week 4 in c/reconstitution · 128 points · 1 year ago
dead space is real and the syringe matters
comment on [Vendor] QST order #15 — Spain, 37 days, no drama, receipts inside in c/maintenancephase · 13 points · 1 year ago
Inter-lab variance on this kind of assay is routinely 1–2 points. Different column, different gradient, different integration.
